Recreational-Water Bacteria Sampling: From Decision Rule to Laboratory Handoff

What to do first

Before visiting the site, obtain the responsible authority's current sampling procedure and decision rule and the selected laboratory's written instructions. Lock the target, approved method, sterile container, sample location and depth basis, timing, required QC, preservation, transport temperature when applicable, holding time, custody, and notification chain. In the field, protect people first, record sanitary conditions, avoid contamination, preserve exact sample identity and time, and deliver within the method-controlled window.

1. Start from the authority decision and work backward

The decision determines indicator, method, stations, time, statistic, and notification speed.

Name the designated or managed use, responsible authority, adopted criterion or beach action procedure, indicator, method, statistic, sampling window, confirmation rule, and public-notification owner. A surveillance sample, source investigation, advisory confirmation, and reopening sample can require different designs.

Record the current public status and verify whether sampling is allowed, required, or directed. The field team documents evidence; it does not independently issue or lift a restriction.

2. Lock laboratory and method requirements before mobilization

The laboratory, not a generic bottle chart, controls the analytical handoff.

3. Place stations and times where the decision applies

Representative design is site- and use-specific, not a universal distance or depth.

Use the sanitary model, recreation area, hydrodynamics, likely exposure zone, inflows/outfalls, known sources, and authority procedure to define stable stations and any targeted event locations. Record coordinates/datum, station version, water-depth and sample-depth basis, shoreline reference, and photo orientation.

Set routine frequency and event triggers for rain, releases, high use, source changes, complaints, blooms, or other program-defined conditions. Do not retrospectively relabel a targeted source sample as representative of the entire recreation area.

4. Complete the safety and sanitary screen before collection

A sample is never more important than exposure control or authority notification.

  • Human illness/exposure report checked and escalated through health procedure
  • Animal illness/exposure report checked and escalated
  • Active sewage, wastewater, chemical release, sheen, or strong odor assessed from a safe location
  • Visible bloom, scum, paint-like streak, or unusual discoloration recorded without contact
  • Water access, current, waves, weather, wildlife, traffic, boat, ice, electrical, and other hazards screened
  • Current signs, barriers, advisory status, and authority direction verified
  • Recent rain, water level/current, outfalls, wildlife, bather load, debris, and photos recorded

5. Preserve sterility, representativeness, and exact time

Small handling errors can create a result that no longer represents the intended sample.

  1. Verify the bottle and label

    Match sample ID, station, target, container, lot/source, analysis, date, and custody record before opening. Keep sterile containers closed until collection.

  2. Collect by the controlled procedure

    Use the exact orientation, depth/basis, facing direction, sampler, gloves, and contamination controls required by the method and site plan. Do not rinse a sterile microbiology container unless the method explicitly requires it.

  3. Protect the sample

    Close immediately, record collection time with UTC offset, preserve and cool exactly as directed, separate from contamination sources, and document deviations.

  4. Complete QC

    Collect required field blanks, replicates, or other QC with unique IDs and the same traceability. Never use a QC sample as a substitute site result.

6. Make the laboratory handoff auditable

Holding time begins at collection, not when the laboratory opens the cooler.

  • Sample IDs and requested analyses match labels, custody, and laboratory order
  • Collection time and UTC offset are unambiguous
  • Preservation/cooling evidence and cooler condition recorded
  • Every transfer includes giver, receiver, date/time, condition, and seal when used
  • Courier delays or temperature/holding deviations reported immediately
  • Laboratory receipt, accession IDs, condition, acceptance/rejection, and analysis start captured
  • Field and laboratory copies reconciled before result interpretation

7. Transfer the complete result, not a naked number

The authority needs result meaning, quality evidence, context, and current status.

The field team may record whether the responsible authority determined a result was within or exceeded its adopted decision rule. The form itself does not calculate legal compliance, authenticate the authority, issue public status, or authorize reopening.

8. Close the event and improve the program

Recurring contamination requires source and system learning, not only repeated sampling.

Archive the raw field, custody, laboratory, result, notification, public-status, and correction records under stable IDs. Review false starts, delays, sample rejection, spatial surprises, illness reports, sanitary sources, communication gaps, and whether stations and triggers still answer the decision.

Use annual sanitary review and after-action review to prioritize source control, infrastructure coordination, additional monitoring, or model development. Retain null, adverse, and inconclusive outcomes.
